Gonzalez hits 3 homers as Rockies rout Astros 13-5

DENVER (AP) ? Carlos Gonzalez's swing appears effortless. The results have been smashing.

Gonzalez extended a power surge by hitting three home runs in his last three at-bats, Michael Cuddyer belted a grand slam and Dexter Fowler also homered, leading the Colorado Rockies past the Houston Astros 13-5 on Wednesday night for their third win in a row.

"All I've been doing is trying to put good swings on the ball," Gonzalez said. "Obviously, I'm not looking for a home run. I'm just looking to hit the ball hard and just make sure it's in the strike zone. I saw three around the strike zone and put a good swing on them. Home runs just happen. I think special nights are just meant to be."

He's been awfully special at the plate lately, going 10 for 16 with five home runs, seven RBIs and six runs scored in his last four games. He has 13 homers on the season.

"His focus at the plate right now, his plate discipline, is as good as I've ever seen it," Rockies manager Jim Tracy said. "He's stepping to another level, that's how dangerous he is."

The game was not without some concern for the Rockies, however. Star shortstop Troy Tulowitzki left the game in the top of the seventh with what the team said was a strained left groin. Tulowitzki appeared a little gimpy as he returned to the dugout after running out an infield groundout in the bottom of the sixth.

"We don't know anything more than just that. We just have to wait and see to what extent," Tracy said of the injury to Tulowitzki, who was scheduled to be re-evaluated on Thursday.

Gonzalez said he just wants to see Tulowitzki get healthy as soon as he can.

"I just want him to be healthy and get right and we all believe he's going to be even better," Gonzalez said. "He's showing signs that he's going to contribute a lot to this team, and I know he's going to do a lot of damage and help the team when he gets back."

Gonzalez did much of the damage against the Astros, breaking a 5-5 tie with a solo shot leading off the fifth off Lucas Harrell (4-4).

"He is a good hitter. If you leave something out over the plate, he's going to hit it hard," Harrell said.

Gonzalez added a two-run drive off reliever Xavier Cedeno as part of a six-run sixth and then had another solo shot, driving the first offering from David Carpenter into the right field seats in the bottom of the eighth inning.

It was Gonzalez's first career three-homer game. He has had four multihomer games this season and eight in his career.

Cuddyer connected in the first inning on the first pitch he saw from Harrell for his seventh career slam and second this season, but it had to survive a replay review by the umpires after a fan reached over the right field railing to catch the ball. The replay showed the ball had cleared the yellow boundary marker atop the right field scoreboard before being caught.

Fowler, who had a game-ending RBI triple among his team record-tying seven hits in Monday's doubleheader sweep of Houston, doubled and scored in the second inning and had a three-run homer off reliever Rhiner Cruz in the sixth for his career-high eighth home run of the season.

"The home runs were the difference," Astros manager Brad Mills said. "Five home runs with the first being a grand slam. We battled back and had a lot of hits. But they won it. They got the five homers."

Harrell, who outdueled the Dodgers' Clayton Kershaw in a 3-1 victory last time out, gave up a career-high nine runs on 10 hits in five innings. He struck out four and walked three.

Christian Friedrich (3-1) allowed five runs on 11 hits in six innings, shaking off a rough third inning in which the Astros scored four runs.

Chris Snyder had a two-run double, Chris Johnson had a sacrifice fly and Justin Maxwell added an RBI single as the Astros pulled into a 5-5 tie. But Houston couldn't sustain the momentum, losing its fifth in a row and its sixth straight to the Rockies at Coors Field.

The Rockies' run total was the highest allowed by the Astros this season, surpassing the previous high of nine, which the Astros had done twice this season, most recently in a 9-7 loss to Colorado in the opener of Monday's doubleheader.

NOTES: Prior to the game, the Rockies designated LHP Jamie Moyer for assignment and recalled pitcher Carlos Torres from Triple-A Colorado Springs. The Rockies have 10 days to trade or release the 49-year-old Moyer, who became the oldest pitcher to win as a starter when he beat the San Diego Padres on April 17. ... Microsoft is also planning to include an optical disk drive in the successor to its Xbox 360 console.

Both Sony's and Microsoft's people declined to comment.

New consoles are expected to be hot topic at the E3 videogame exposition in Los Angeles next week, though the only formal announcement expected is a successor for Nintendo 's Wii, the new Wii U.

Facebook has its eyes on Face.com, reports say

Reports in the tech press Tuesday said that Facebook, whose stock is still reeling from the aftershocks of a bungled IPO, is looking to acquire Israel-based face recognition technology company Face.com for around $80 million to $100 million.

The Los Angeles Times reported that rumors of the deal for the startup surfaced in the Israeli press and then were quickly picked up by tech reporters. Several of the reports said that when they contacted Face.com CEO Gil Hirsch, he responded by saying he had no news to share.

Shares of Facebook (FB) were down about 7 percent on Tuesday to just below $30 on a day when most market indices were higher. The fiber speed-up is being attributed to a flood of Internet video and cloud backups, both of which get downright reasonable at 300Mbps; Verizon figures that both a 2-hour HD movie download and a 10GB backup will finish in 22 minutes or less. Whether or not pricing will have changed will have to wait until the speed upgrades take effect in June. Somehow, we can't imagine a drop anytime soon in the $200 monthly outlay for the top tier.

Sixteen-year-old Shouryya Ray, a student from Dresden who was born in Calcutta, submitted a paper proposing analytical solutions to two problems in particle dynamics.

A research paper that claims to fill in a gap in Isaac Newton's formulas for the physics of falling objects has drawn worldwide attention to a 16-year-old student in Germany, but physicists are reserving judgment until they've seen the proof.

The focus of the buzz is Shouryya Ray, an Indian-born student who won second prize this month in the math and informatics category for Germany's Jugend Forscht student science competition. Ray tackled a couple of longstanding puzzlers for physics students: How do you account for air resistance in calculating the trajectory of ball thrown out at an angle? And precisely how does a ball thrown against the wall rebound?


The first question relates to Newton's law of universal gravitation: In his Principia Mathematica, published in 1687, Newton laid out how a gravitational field would affect a thrown object ??but he didn't account for the effect of air resistance. Through the centuries, physicists have used numerical approximations to take drag into account, and when computers come into play, those approximations can be incredibly precise. But Ray said he wanted to come up with a set of formulas that could calculate the effect directly, even though his instructors said that had never been done.

"I asked myself: Why can't it work?" he told the German newspaper Die Welt.

That's what Ray tried to do in his prize-winning paper, titled "Analytical?Solution of Two?Fundamental?Unsolved?Problems of?Particle Dynamics" ("Analytische L?sung von zwei ungel?sten fundamentalen Partikeldynamikproblemen"). In addition to the falling-ball problem, Ray took on a puzzler of more recent vintage, having to do with the description of a particle's collision with a wall, as described by 19th-century theory. But it was the "kid-trumps-Newton" angle that really stirred up a buzz.

Die Welt's report came early in the game: The Daily Mail and?The Sunday Times of London picked up the story, adding to the sensation. The idea that a teenager could figure out something that Newton didn't is irresistible ? particularly when the teen is an immigrant from Calcutta who says he's no genius. But the story just sparked more questions among inquiring minds in such online hangouts as?Physics Forum,?Slashdot?and Reddit: What exactly did Ray do? And were these problems really such mysteries to solve?

That's a challenge, because Ray's paper was a school project submitted for a contest, and thus not subject to the publication process and peer review that professional work typically goes through. For that reason, the experts are reluctant to weigh in.

"This story seems rather suspicious," Richard Fitzpatrick, a physicist at the University of Texas in Austin, told me in an email. "None of the news reports give any details of the calculation. None of the people who hailed Shouryya Ray as a genius are scientists, and none of?them give the impression that they have seen the calculation in question. It is impossible to gauge the scientific merit of the calculation until it is made public."

Syracuse University physicist Simon Catterall said in an email that calculating the trajectories of falling objects hadn't been seen as a particularly grand puzzle of physics. "The background given in the article seems genuine enough, so it may indeed be true, but I haven't heard anything about a new solution to a Newtonian problem on the grapevine," he told me.

Based on what's come out about the work so far, the consensus seems to be that Ray has done amazing work for his age ? and if he had to choose between his passion for science and his passion for soccer, he'd be well-advised to pick math and physics.?His paper putting forth an "analytical solution to two fundamental unsolved problems" may not be the breakthrough that some of the reports have made it out to be, but that doesn't take anything away from the teenager's achievement.

"What Ray has worked out, almost certainly independently, would definitely put him in the 99th percentile amongst his peers and maybe even more," one Redditor observed.

By the way, the first-place winner in the math and informatics category, Julius Kunze, wrote a paper on relativistic ray tracing. But that's a different story...?

Update for 5 p.m. ET: Other experts on Newtonian physics have replied to my follow-up queries via email:

Oxford University physicist James Binney: "Doesn't sound too interesting to me. The resistance of air to the ball won't be susceptible to simple analytic formulae ??if the ball is of ordinary size, [greater than a centimeter] radius ??the flow around it will be in the high Reynolds-number regime and involve a thin boundary layer. Such flows were extensively studied from the last part of the 19th century, so it's true that they lie beyond Newton's knowledge. A good approximation will be to take the drag force as pi r^2 rho v^2, where r is the radius of the ball, v its speed and rho the density of air. I'm unaware of a puzzle regarding bouncing balls. In detail the bounce will depend on the physical properties of the ball ??as any squash player knows. Usually one adopts a coefficient of restitution.?To be impressed we need to know details."

University of Bristol physicist Michael Berry: "Without seeing the details of what Ray has claimed, it's impossible to comment intelligently. It depends crucially on how he has modeled the air resistance. But a falling body with air resistance (however modeled) is hardly a 'fundamental unsolved problem,' as he seems to think. There's a powerful aroma of hype."??

It took Earth ten million years to recover from greatest mass extinction

ScienceDaily (May 27, 2012) ? It took some 10 million years for Earth to recover from the greatest mass extinction of all time, latest research has revealed.

Life was nearly wiped out 250 million years ago, with only 10 per cent of plants and animals surviving. It is currently much debated how life recovered from this cataclysm, whether quickly or slowly.

Recent evidence for a rapid bounce-back is evaluated in a new review article by Dr Zhong-Qiang Chen, from the China University of Geosciences in Wuhan, and Professor Michael Benton from the University of Bristol. They find that recovery from the crisis lasted some 10 million years, as explained May 27 in Nature Geoscience.

There were apparently two reasons for the delay, the sheer intensity of the crisis, and continuing grim conditions on Earth after the first wave of extinction.

The end-Permian crisis, by far the most dramatic biological crisis to affect life on Earth, was triggered by a number of physical environmental shocks -- global warming, acid rain, ocean acidification and ocean anoxia. These were enough to kill off 90 per cent of living things on land and in the sea.

Dr Chen said: "It is hard to imagine how so much of life could have been killed, but there is no doubt from some of the fantastic rock sections in China and elsewhere round the world that this was the biggest crisis ever faced by life."

Current research shows that the grim conditions continued in bursts for some five to six million years after the initial crisis, with repeated carbon and oxygen crises, warming and other ill effects.

Some groups of animals on the sea and land did recover quickly and began to rebuild their ecosystems, but they suffered further setbacks. Life had not really recovered in these early phases because permanent ecosystems were not established.

Professor Benton, Professor of Vertebrate Palaeontology at the University of Bristol, said: "Life seemed to be getting back to normal when another crisis hit and set it back again. The carbon crises were repeated many times, and then finally conditions became normal again after five million years or so."

Finally, after the environmental crises ceased to be so severe, more complex ecosystems emerged. In the sea, new groups, such as ancestral crabs and lobsters, as well as the first marine reptiles, came on the scene, and they formed the basis of future modern-style ecosystems.

Professor Benton added: "We often see mass extinctions as entirely negative but in this most devastating case, life did recover, after many millions of years, and new groups emerged. The event had re-set evolution. However, the causes of the killing -- global warming, acid rain, ocean acidification -- sound eerily familiar to us today. Perhaps we can learn something from these ancient events."

That sentiment is fueling Canada's most sustained student demonstrations ever. It has been anything but quiet.

Some 150,000 students in more than a dozen Quebec colleges and universities have been on strike since February to protest the provincial government's plan to raise tuition fees. Street protests in Montreal have ended in clashes with police and mass arrests.

A strict new law designed to stop the demonstrations has only broadened the movement to include separatists and Occupy protesters, and triggered a wider debate over public freedoms. The students are threatening to persevere through the summer, just when the city traditionally awakens from its dark and frigid winter for jazz and comedy festivals that draw in millions of dollars in tourist revenue.

The French-speaking province's average undergraduate tuition ? $2,519 a year ? is the lowest in Canada, and the proposed hike? $254 per year over seven years ? is tiny by U.S. standards. But opponents consider the raise an affront to the philosophy of the 1960s reforms that set Quebec apart not only from its U.S. neighbor but from the rest of Canada.

"The whole consensus around education was built around the Quiet Revolution," said Pierre Martin, a political science professor at the Universite de Montreal. "That consensus would tend toward a tuition-free model in the future. That was a promise."

As a result, he said, Quebecers don't compare their tuition rates to those in the U.S. or English-speaking Canada, but to those in European countries where higher education is free.

Quebec Premier Jean Charest, who has vowed to shake up the debt-ridden province's finances since he was elected nearly a decade ago, has refused to cave.

More than 2,500 students have been arrested since the demonstrations began more than 100 days ago, including nearly 700 this past Wednesday alone. The total is five times the arrests during a period in the 1970s when soldiers were deployed to the streets in Quebec because of a spate of terrorism by a group demanding independence from Canada.

The tuition hike is part of a broader effort to shift Quebec's fiscal burden away from taxpayers ? the province has some of the steepest personal income taxes in North America and the highest per-capita debt in Canada? and onto the shoulders of each person who uses a service.

"Every citizen has to do their part," Quebec Finance Minister Raymond Bachand told The Associated Press in a telephone interview. "This is the 21st century."

Bachand said the students want more money in their pocket to the detriment of others. He noted that the government has expanded student grants so that middle and lower income students can more easily afford the increase.

"We're talking about 50 cents a day so basically it's moved from a question of tuition fees to a question of a social movement like you've seen in other parts of the world," Bachand said. "We're not used to this as Canadians. We're used to sitting down, disagreeing, negotiating and coming to an agreement."

In an effort to restore peace, Charest's government passed emergency legislation on May 18 restricting protests and closing striking campuses until August. The law requires that police be informed eight hours before a protest begins, including details on the route of any demonstration of 50 or more people. It also prohibits demonstrations within 50 meters (165 feet) of a college and declares that anyone who incites or helps another person break the new protest regulations can be fined.?

The new law has "changed the discourse now and introduced a new element into the debate, and that is the question of rights," said Bruce Hicks, a political scientist at Concordia University. The students have challenged the law in court.

Provincial Public Safety Minister Robert Dutil said Quebec cities are simply joining others that already have tough rules for organizing protests, including Los Angeles and New York.

"Law 78" has inspired a particularly cacophonous form of protest: from 8 p.m. to 8:15 p.m., people around Quebec emerge onto sidewalks and balconies to bang pots and pans.

Across the street, pizza parlor owner Naeem Ahmed shook his head when he recalled the sight of an open fire hydrant gushing water into his business and a bonfire lit dangerously close by.

Ahmed, 37, has had a front row seat to more than 30 straight nights of protests. Confrontations between helmeted police and students have scared customers away. Students sometimes seek shelter in his establishment to escape police sweeps.

"For businesses it has been really, really horrible," Ahmed said, taking a break with some of his employees to look at cellphone footage of a recent protest. One of his employees joined the pot-bangers, clanging two trash lids together.

"Police come and ask everybody to leave," he said.

Still, he sympathizes with the students. "If you go to any country, when the students hit the streets, you have to listen to them," he said. "When they come out you have to accept it."

The student groups and the government have announced their intention to return to negotiations in the next days, with Quebec Education Minister Michelle Courchesne saying she expects a "very, very important" session following positive discussions over the phone.

But the two sides seem far apart. Student leader Gabriel Nadeau-Dubois wants to propose a long-term plan that would essentially fulfill the promise of the Quiet Revolution: scrapping university tuition fees altogether within five years and funding that by increasing taxes on financial institutions.

That is the opposite of what the government wants.

"Sacred cows only exist in India," the finance minister has repeatedly said.

Finding Your Voice | GeekGirlCon

Hey guys, Shubz here! Anyone on staff will tell you that when I?m in the room, you?re more than likely to hear me (and laugh). However, it wasn?t always the case. I never had a problem speaking out or speaking my mind, but it never seemed like it was for myself. You see, I usually am speaking on behalf of something, whether it be my job or my family. It wasn?t until a year ago that I discovered that I CAN have my own voice ? literally and figuratively.

Here's a re-enactment of how things went for me in ballet class.

Time travel back 20+ years, and you would have found me in a dance class, learning the delicate dance form known as ballet. While ballet became a gateway to other forms like jazz, contemporary, and (my favorite) hip-hop, I hid behind my choreography. I chose to use movement as a way for people to see and experience my view without ever having to say a thing. I loved it. Every kick and every step had a purpose and a meaning. I thought there was nothing else that I could use as a platform to fully express who I was.

This is where magic gets written. Behold the notebook!

Let?s warp speed it back to the near present; my body has succumbed to the wear and tear of an active childhood and early adulthood. I no longer am active as a dancer, and it left quite a void in my life that I didn?t have an outlet with which I could create. I found writing by chance, a dear friend involved in local theatre heard me rap Sir Mix-A-Lot?s ?Baby Got Back? and insisted I begin to write. Eventually, I meet my husband, Aaron, who introduced me to this genre of Nerdcore and a community, (NerdcoreNow), where others are producing music.

When Aaron and I are not rapping, we are practicing our gloriously awesome poses.

I wrote, I recorded, and I performed. While I was met by many who enjoyed my music, I had my setbacks. There were folks that dismissed women rapping and others that didn?t like what I had to rap about. However, I couldn?t quit. I have never been the person to throw in the towel ? not in dance, and certainly not in this art form that I had just begun to play with. My perseverance paid off. I was awarded with NerdcoreNow?s Best Newcomer Award for 2011, and I got the respect that I thought I wouldn?t get by just writing and speaking into a mic. The acknowledgement is something that I refuse to rest my laurels on; now I?m working on an album!

I always wonder what would have happened if I didn?t take up my friend?s encouragement to write. What would I end up doing for a creative outlet? WOULD I have one? What?s great is that I don?t have to answer those questions because I just got brave enough to stretch myself out to try something that I thought I would never do. Sure, I?ve only just begun, but if this is how my start has panned out, I can?t wait to experience the journey that lies ahead!

Group: S.Sudan troops kill, torture minority tribe

JUBA, South Sudan (AP) ? South Sudan military forces tasked with carrying out a disarmament campaign among feuding ethnic groups are raping, torturing and killing members of a minority community, community leaders and aid workers say.

The disarmament campaign follows two outbreaks of violence linked to cattle raids between the Murle and Lou Nuer tribes over the last year in the remote state of Jonglei. Hundreds ? and likely more than 1,000 ? people were killed in the two clashes.

In an effort to stem future violence, the Sudan People's Liberation Army, South Sudan's military, embarked on a disarmament campaign in March in which more than 10,000 weapons have been taken so far, said military spokesman Col. Philip Aguer.

But aid groups and community members say the SPLA is killing and torturing members of the Murle, a tribe reviled by many other South Sudanese, while carrying out the disarmament campaign. Alleged abuses include simulated drownings, tying up young men to trees and beating them, and widespread rape against women.

The Murle appear to be ostracized by most other tribes in South Sudan in part because they received military support from leaders in Khartoum, Sudan, when the south and north battled in a two-decade civil war. The Murle also have a reputation for carrying out child abductions from other tribes and of conducting cattle raids that can results in hundreds of human deaths and tens of thousands of stolen cattle.

Allegations of abuse during the disarmament campaign are being investigated by military lawyers, Aguer said.

A state government report dated March 30 and obtained by The Associated Press listed some of the deaths: James Kengen Logidang was killed at home while sitting in a chair. Korok Manyngar Kengen, 15, was killed as his cattle were being stolen by SPLA soldiers. "The boy didn't have a gun," the report said.

The aid group Medecins Sans Frontieres, or Doctors Without Borders, said it has treated 30 patients since mid-March who suffered injuries from the disarmament campaign. Two have since died. Three patients had gunshot wounds and 26 had trauma injuries from beatings, the group said.

Aid groups contend that such numbers are underreported because most of the attacks take place in remote areas.

The disarmament campaign began as a voluntary handing over of guns, and then was to move into a compulsory phase, though military leaders have since said the voluntary phase would continue. Aid groups question whether the process is truly voluntary.

Peter Guzulu, a member of the Murle community, said at least 10 people have been killed "for no apparent reason." An 11th death took place against a male running from soldiers with a rifle in hand, he said.

Guzulu was the chairman of the government-run Jonglei Human Rights Commission from September 2010 until mid-March, when he said he was dismissed by the governor but wasn't told why. Guzulu believes he was removed "so I didn't talk to journalists or write reports."

"People are being beat in the towns and villages. They are told to confess to having guns. Women are beaten. Some are raped. When is this torture going to end? That is the question people ask all the time," he said.

Aguer said all communities in Jonglei are being disarmed. He said the Murle have the most weapons and that some members are heading into Ethiopia to keep their AK-47 assault rifles.

A report published last month by five aid groups, including Washington D.C.-based Pact and the South Sudan Law Society, said the current disarmament campaign ? the fifth in Jonglei in the last six years ? risks perpetuating a cycle of violence that has seen thousands killed and hundreds of thousands displaced in recent years. If the Murle believe they are being forcibly targeted in the campaign, the report said, they will re-arm and seek revenge.

The report urges the U.N. to prioritize the protection of civilians.

A statement this month from the U.N.'s top representative in South Sudan, Hilde Johnson, commended the disarmament effort, but critics of the campaign say the U.N. mission in South Sudan is ignoring evidence of abuse and torture.

The U.N. said in April that a peaceful disarmament campaign that respects human rights is vital to ending the cyclical violence and noted that while cases of abuse and sexual violence have been reported they do not appear to be part of a systematic campaign.

Kouider Zerrouk, a spokesman for the U.N. mission, said the U.N. has no way of knowing how many cases of abuse have occurred.

"These human rights violations are serious and are a concern for us," Zerrouk said. "But if we look into the whole process covering the whole region as big as Bangladesh, yes, generally it went in a peaceful manner in the sense that there was no outbreak of major clashes between communities."

Aguer said the disarmament campaign would continue for the rest of the year.

Judy McCallum, the former head in South Sudan of the aid group Pact, said the Murle are "basically under siege" by the government and military. She added that Murle youth are "out of control" ? a reference to cattle-raiding attacks ? but have no path to improve their lives.

Aid groups, McCallum said, are reporting systemic rape and drownsings. "Everyone I talk to say women are being raped constantly. So there's no incentive to give up their guns," she said. She said there is impunity for violence against the Murle.

McCallum said the U.N. is overemphasizing the success of the disarmament campaign and does not appear to be adequately concerned with the protection of civilians.

Minority Rights Group International on Thursday released new rankings of peoples under threat. South Sudan ? which broke away from Sudan last year to become the world's newest nation ? ranked in the top 10 because of the Murle-Lou Nuer violence.

In December and January, members of the Lou Nuer community attacked the Murle, likely killing hundreds of people. That attack came in retaliation for an August attack by the Murle that killed at least 600.

Northern California man finds giant oyster

(AP) ? A northern California man who's been searching for years for a record-sized oyster appears to have found one.

The Eureka Times-Standard reports (http://bit.ly/JAp102 ) Richard Mesce found the 13-inch-long, 6-inch-wide mollusk in the muddy bottom of Humboldt Bay. It's estimated at 80 years old.

The retired pilot took the oyster to a Humboldt State University oceanographer for an official measurement, a requirement of the Guinness World Records' application.

The current record-holding oyster measures 12 inches long and 5 ? inches wide.

Mesce started searching in 2006, when he happened upon a large oyster in the area. Paddling in his kayak, he continued his quest when the tide was right.

If the oyster is confirmed the largest, Humboldt State University has agreed to keep the mollusk in its aquarium, where it can continue to grow.

Patients' skin cells turned into heart muscle cells to repair their damaged hearts

ScienceDaily (May 22, 2012) ? For the first time scientists have succeeded in taking skin cells from heart failure patients and reprogramming them to transform into healthy, new heart muscle cells that are capable of integrating with existing heart tissue.

The research, which is published online May 22 in the European Heart Journal, opens up the prospect of treating heart failure patients with their own, human-induced pluripotent stem cells (hiPSCs) to repair their damaged hearts. As the reprogrammed cells would be derived from the patients themselves, this could avoid the problem of the patients' immune systems rejecting the cells as "foreign." However, the researchers warn that there are a number of obstacles to overcome before it would be possible to use hiPSCs in humans in this way, and it could take at least five to ten years before clinical trials could start.

Recent advances in stem cell biology and tissue engineering have enabled researchers to consider ways of restoring and repairing damaged heart muscle with new cells, but a major problem has been the lack of good sources of human heart muscle cells and the problem of rejection by the immune system. Recent studies have shown that it is possible to derive hiPSCs from young and healthy people and that these are capable of transforming into heart cells. However, it has not been shown that hiPSCs could be obtained from elderly and diseased patients. In addition, until now researchers have not been able to show that heart cells created from hiPSCs could integrate with existing heart tissue.

Professor Lior Gepstein, Professor of Medicine (Cardiology) and Physiology at the Sohnis Research Laboratory for Cardiac Electrophysiology and Regenerative Medicine, Technion-Israel Institute of Technology and Rambam Medical Center in Haifa, Israel, who led the research, said: "What is new and exciting about our research is that we have shown that it's possible to take skin cells from an elderly patient with advanced heart failure and end up with his own beating cells in a laboratory dish that are healthy and young -- the equivalent to the stage of his heart cells when he was just born."

Ms Limor Zwi-Dantsis, who is a PhD student in the Sohnis Research Laboratory, Prof Gepstein and their colleagues took skin cells from two male heart failure patients (aged 51 and 61) and reprogrammed them by delivering three genes or "transcription factors" (Sox2, Klf4 and Oct4), followed by a small molecule called valproic acid, to the cell nucleus. Crucially, this reprogramming cocktail did not include a transcription factor called c-Myc, which has been used for creating stem cells but which is a known cancer-causing gene.

"One of the obstacles to using hiPSCs clinically in humans is the potential for the cells to develop out of control and become tumours," explained Prof Gepstein. "This potential risk may stem from several reasons, including the oncogenic factor c-Myc, and the random integration into the cell's DNA of the virus that is used to carry the transcription factors -- a process known as insertional oncogenesis."

The researchers also used an alternative strategy that involved a virus that delivered reprogramming information to the cell nucleus but which was capable of being removed afterwards so as to avoid insertional oncogenesis.

The resulting hiPSCs were able to differentiate to become heart muscle cells (cardiomyocytes) just as effectively as hiPSCs that had been developed from healthy, young volunteers who acted as controls for this study. Then the researchers were able to make the cardiomyocytes develop into heart muscle tissue, which they cultured together with pre-existing cardiac tissue. Within 24-48 hours the tissues were beating together. "The tissue was behaving like a tiny microscopic cardiac tissue composed of approximately 1000 cells in each beating area," said Prof Gepstein.

Finally, the new tissue was transplanted into healthy rat hearts and the researchers found that the grafted tissue started to establish connections with the cells in the host tissue.

"In this study we have shown for the first time that it's possible to establish hiPSCs from heart failure patients -- who represent the target patient population for future cell therapy strategies using these cells -- and coax them to differentiate into heart muscle cells that can integrate with host cardiac tissue," said Prof Gepstein.

"We hope that hiPSCs derived cardiomyocytes will not be rejected following transplantation into the same patients from which they were derived. Whether this will be the case or not is the focus of active investigation. One of the obstacles in dealing with this issue is that, at this stage, we can only transplant human cells into animal models and so we have to treat the animals with immunosuppressive drugs so the cells won't be rejected."

Much research has to be conducted before these results could be translated into treatment for heart failure patients in the clinic. "There are several obstacles to clinical translation," said Prof Gepstein. "These include: scaling up to derive a clinically relevant number of cells; developing transplantation strategies that will increase cell graft survival, maturation, integration and regenerative potential; developing safe procedures to eliminate the risks for causing cancer or problems with the heart's normal rhythm; further tests in animals; and large industry funding since it is likely to be a very expensive endeavour. I assume it will take at least five to ten years to clinical trials if one can overcome these problems."

Prof Gepstein and his colleagues will be carrying out further research into some of these areas, including evaluating using hiPSCs in cell therapy and tissue engineering strategies for repairing damaged hearts in various animal models, investigating inherited cardiac disorders, and drug development and testing.
